I would definitely say J.K. Rowling's speech for the 2011 Harvard Commencement - absolutely outstanding!

I also love the speech attributed to Kurt Vonnegut but I think that was by another author actually.

To prican11: Are those commencement speeches available on YouTube? I'd be very interested in seeing them. I am a fan of both Vonnegut and Seuss.

I enjoyed Neil Gaiman's "Make Good Art" commencement speech. I'll have to look up Rowling's speech now.

She isn't strictly an author, but maybe because of Oprah's Book Club, we can include her? Oprah gave a really great speech at Stanford a few years back. She has a very clear outline and some solid advice for new graduates. We watched it in one of my classes right before graduating high school, and apparently it made an impression because I remember it.
